Polysorbate 80
Polysorbate 80 is an emulsifier that helps oil and water stay mixed in foods such as frozen desserts, sauces, and dressings. It is made from sorbitol and fatty acids, with added oxyethylene units, and differs from similarly named polysorbates and sorbitan esters.

The review considers repeated experimental gut effects, incomplete long-term human evidence and dose-dependent findings, balanced against a small human trial without clear inflammatory changes and a newer animal study with largely reassuring results.

Polysorbate 80 helps maintain texture and disperse ingredients. It has more direct gut-health research than several related emulsifiers, but most evidence comes from animals and laboratory systems. Mouse studies have found changes in gut bacteria, inflammation, metabolism, and susceptibility to disease, including effects after maternal exposure. Some experiments found changes at lower concentrations, so the evidence cannot all be dismissed as extreme dosing.
However, a newer study using smaller amounts in an oil emulsion found no general increase in inflammation or intestinal permeability. A small human trial tested several emulsifiers for four weeks and found no clear differences in measured inflammation or metabolic markers; its microbial-metabolite findings need careful interpretation by additive. A larger cross-sectional study linked P80-associated metabolites with Crohn disease activity, but could not establish whether food exposure preceded disease or distinguish all medication and related-polysorbate sources; short trials cannot settle long-term effects. Older animal tests also included gastrointestinal injury and equivocal tumor findings at high doses, which regulators weighed when setting intake limits.
In 2026, JECFA reaffirmed a daily limit for polysorbates together and found current Codex uses acceptable, while requesting better impurity data. The review considers unresolved gut-health signals and exposure differences, rather than demonstrated disease from ordinary permitted food intake.
